Second-placed Osaka, who was born in Japan and grew up in the USA, struggled with her otherwise reliable basic strokes.
The 42nd Vondrousova produced a string of drop shot winners and other sly shots that got Osaka out of their comfort zone.
Osaka won their first two games in straight sets after a two-month mental hiatus. But the conditions were different on Tuesday with the roof closed because it was raining outside.
